- Peptide ELISA: antibody detection limit dilution 1: 128000. Western blot: Preliminary experiments gave an approx 70-75 kDa band in Human BoneMarrow, Duodenum and Skin lysates after 0.01 μg/mL antibody staining. Please note thatcurrently we cannot find an explanation in the literature for the band we observe given thecalculated size of 57.5 kDa according to NP_001354.1. The 70-75 kDa band was successfullyblocked by incubation with the immunizing peptide. - DKC1 (Dyskeratosis congenita 1) is a nucleolar protein, altered in dyskeratosis congenita, which carries out two separate functions, both fundamental for proliferating cells. One function is the pseudouridylation of ribosomal RNA (rRNA) molecules, necessary for their processing, and the other is the stabilization of the telomerase RNA component, necessary for telomerase activity. It is thought to be the catalytic subunit of the H/ACA small nucleolar ribonucleoprotein (H/ACA snoRNP) complex, which catalyzes pseudouridylation of rRNA. Each rRNA can contain up to 100 pseudouridine ('psi') residues, which may serve to stabilize the conformation of rRNAs. DKC1 is also required for correct processing or intranuclear trafficking of TERC, the RNA component of the telomerase reverse transcriptase (TERT) holoenzyme.
