- Background: In addition to its possible functions as a facilitator of Myc activated transcription, PAM (protein associated with Myc) has also been shown to play a role in mTOR signaling. PAM functions as an E3 ubiquitin ligase for tuberin (TSC2), a negative regulator of the mTOR pathway. Additionally PAM has been shown to function as a guanosine exchange factor (GEF) for Rheb and to facilitate sphingosine-1-phosphate signaling in the mTOR pathway.
