This competitive ELISA uses the microtiter plate format. The antigen is bound to the solid phase of the microtiter plate. The acylated standards, controls and samples and the solid phase bound analyte compete for a fixed number of antiserum binding sites. When the system is in equilibrium, free antigen and free antigenantiserum complexes are removed by washing. The antibody bound to the solid phase is detected by an antirabbit IgGperoxidase conjugate using TMB as a substrate. The reaction is monitored at 450 nm. Determination of unknown samples is achieved by comparing their absorbance with a reference curve prepared with known standards.

Gamma-Aminobutyric acid (GABA) is the chief inhibitory neurotransmitter in the mammalian central nervous system. It plays a role in regulating neuronal excitability throughout the nervous system. In humans, GABA is also directly responsible for the regulation of muscle tone. In vertebrates, GABA acts at inhibitory synapses in the brain by binding to specific transmembrane receptors in the plasma membrane of both pre and postsynaptic neuronal processes.
